摘要:

Microfluidic lab-on-a-chip devices provide a flexible and cost-effective platform to implement chemical, bio-chemical, and bio-analytical processes. As a result, they are increasingly used in biomedical research, in particular in the form of perfusion culture systems for cells, cell aggregates, tissues, and small organisms. Most of these assays rely on end-point readout of the experimental outcome via fluorescence assays. At the same time, Nuclear Magnetic Resonance (NMR) spectroscopy is capable of providing detailed insight into chemical and transport processes in real time, in a completely non-invasive manner.

Over the past few years, our group have developed a dedicated NMR probe assembly that accommodates generic microfluidic devices with a sampling volume between 2 and 5 µl. These detectors reach a mass limit of detection of about 1 nMol/sqrt(s), and thus allow quantification of mM concentrations with relative ease. In the present presentation, the architecture and performance of these detectors will be discussed, and some examples of applications to tissue slice culture, metabolomic monitoring of cell cultures, and to chemical kinetic studies will be discussed. Some recent results on microfluidic hyperpolarised NMR using para-hydrogen induced polarisation will also be presented.
