报告摘要:
We have developed a hairpin DNA assembly technology that enables cancer-selective immune activation to induce cytotoxicity. The designed artificial DNA hairpins assemble into long nicked double-stranded DNA triggered by intracellular microRNA-21 (miR-21), which is overexpressed in various types of cancer cells. The products from the hairpin DNA assembly selectively kill miR-21-abundant cancer cells in vitro and in vivo based on innate immune activation. Our approach is the first to allow selective oncolysis derived from intracellular DNA self-assembly, providing a powerful therapeutic modality to treat cancer. We will also introduce the functions and applications of the fluorescent nucleic acids we previously developed.
报告人简介:
Akimitsu Okamoto is a professor in the Department of Chemistry and Biotechnology at the School of Engineering, the University of Tokyo. Professor Okamoto earned his Bachelor's, Master's, and Doctoral degrees of Engineering from Kyoto University, Japan, and conducted postdoctoral research at the Massachusetts Institute of Technology, USA. Prior to joining the University of Tokyo in 2012, he held positions at RIKEN and Kyoto University in Japan. His research interests span synthetic chemistry, bioorganic chemistry, and nucleic acid chemistry, with his work published in leading journals such as Nature Chemical Biology, Journal of the American Chemical Society, and Angewandte Chemie International Edition. Professor Okamoto has received numerous accolades, including the Organic & Biomolecular Chemistry Lecture Award from the Royal Society of Chemistry, the Young Scientists' Prize from the Ministry of Education, Culture, Sports, Science and Technology of Japan, and the Young Chemist Award from the Chemical Society of Japan. He also serves as a board member for the Chemical Society of Japan and the Japanese Society of Nucleic Acids Chemistry, and as a director of TKG Therapeutics.
